NPC/HK1

NPC/HK1（亦稱為 HK-1 或 HK1）是一種從一名 58 歲中國男性患者的鼻咽部建立的人類鼻咽癌（NPC）細胞系。 NPC/HK1 以黏附性上皮單層形式生長，倍增時間約為 28 ± 2 小時。NPC/HK1 是廣泛用於研究未分化型鼻咽癌的模型，此種惡性腫瘤與愛潑斯坦-巴爾病毒（EBV）感染密切相關，且在東南亞人群中尤為常見。 該細胞系於香港建立，提供了一種 EBV 陰性或低 EBV 含量的模型，可與 EBV 陽性鼻咽癌細胞系互補，用於研究鼻咽癌生物學中與 EBV 無關的方面。

NPC/HK1 適用於鼻咽癌生物學、上皮-間質轉化、侵襲與轉移、EBV 生物學（與 EBV 陽性細胞系進行感染與潛伏期比較）、EGFR 訊號傳導途徑，以及化療／放射治療敏感性的研究。 該細胞系用於免疫功能低下宿主的異種移植模型，並作為鼻咽癌細胞系組合的一部分，用於進行藥理學和分子學的比較研究。其源自中國男性捐贈者的背景，反映了鼻咽癌風險最高的族群，對於EBV流行病學及鼻咽癌免疫基因組學的研究具有重要意義。

37°C，5% CO_₂，加濕環境。

1. Confirm that the vial remains deeply frozen upon delivery, as cells are shipped on dry ice to maintain optimal temperatures during transit.

2. Upon receipt, either store the cryovial immediately at temperatures below -150°C to ensure the preservation of cellular integrity, or proceed to step 3 if immediate culturing is required.

3. For immediate culturing, swiftly thaw the vial by immersing it in a 37°C water bath with clean water and an antimicrobial agent, agitating gently for 40-60 seconds until a small ice clump remains.

4. Perform all subsequent steps under sterile conditions in a flow hood, disinfecting the cryovial with 70% ethanol before opening.

5. Carefully open the disinfected vial and transfer the cell suspension into a 15 ml centrifuge tube containing 8 ml of room-temperature culture medium, mixing gently.

6. Centrifuge the mixture at 200 x g for 5 minutes, carefully discard the supernatant containing freezing medium.

7. Follow the procedure described under Post-Thaw Recovery

若需長期保存，請將試管置於溫度約為 −150 至 −196 °C 的氣相液氮中。僅在轉移至液氮之前，可將試管暫時存放於 −80 °C 環境中，作為短期過渡步驟。
