해동 및 배양

  1. Confirm that the vial remains deeply frozen upon delivery, as cells are shipped on dry ice to maintain optimal temperatures during transit.

  2. Upon receipt, either store the cryovial immediately at temperatures below -150°C to ensure the preservation of cellular integrity, or proceed to step 3 if immediate culturing is required.

  3. For immediate culturing, swiftly thaw the vial by immersing it in a 37°C water bath with clean water and an antimicrobial agent, agitating gently for 40-60 seconds until a small ice clump remains.

  4. Perform all subsequent steps under sterile conditions in a flow hood, disinfecting the cryovial with 70% ethanol before opening.

  5. Carefully open the disinfected vial and transfer the cell suspension into a 15 ml centrifuge tube containing 8 ml of room-temperature culture medium, mixing gently.

  6. Centrifuge the mixture at 300 x g for 3 minutes to separate the cells and carefully discard the supernatant containing residual freezing medium.

  7. Gently resuspend the cell pellet in 10 ml of fresh culture medium. For adherent cells, divide the suspension between two T25 culture flasks; for suspension cultures, transfer all the medium into one T25 flask to promote effective cell interaction and growth.

  8. Adhere to established subculture protocols for continued growth and maintenance of the cell line, ensuring reliable experimental outcomes.

냉동 보존 배지로는 해동 후 충분한 생존력을 위해 완전 성장 배지(FBS 포함) + 10% DMSO를 사용하거나, 최적화된 삼투 보호제 및 대사 안정제가 포함된 CM-1(Cytion 카탈로그 번호 800100)을 사용하여 회복력을 높이고 냉동으로 인한 스트레스를 줄입니다.
보관 조건

장기 보존을 위해서는 바이알을 약 -150°C에서 -196°C의 증기상 액체 질소에 넣으세요. 80°C에서 보관하는 것은 액체 질소로 옮기기 전 짧은 중간 단계로만 허용됩니다.

냉동 보존 세포주는 운송 중 약 -78°C를 유지할 수 있는 충분한 냉매가 포함된 검증된 단열 포장에 드라이아이스를 넣어 배송됩니다. 수령 즉시 용기를 검사하고 바이알을 지체 없이 적절한 보관 장소로 옮깁니다.
