Hep-G2/C3A

Hep-G2/C3A（常簡稱為 C3A）是一種從一名 15 歲男性患者的肝腫瘤（肝母細胞瘤）中建立的人類（智人，Homo sapiens）細胞系。 它是廣為人知的 Hep-G2 細胞系的克隆亞系，因此源自惡性肝組織。作為源自肝臟的癌症細胞系，Hep-G2/C3A 在體外呈現持續生長，並被廣泛用作穩定且可重複的人類肝細胞模型。

憑藉其強勁的生長特性及保留了關鍵的肝臟特異性功能，Hep-G2/C3A 被廣泛應用於多個研究領域。在藥理學與毒理學領域中，它作為研究藥物代謝與肝毒性的體外模型尤為常見，其中包含可提升人類肝毒性預測精準度的 3D 球體培養模式。 在癌症研究中，Hep-G2/C3A 作為肝腫瘤（肝細胞癌／肝母細胞瘤）的模型，並支援在受控的實驗室條件下測試治療策略。 由於其功能與原代肝細胞相似（例如：產生血漿蛋白及進行活性代謝），該細胞系亦被應用於生物人工肝支援系統的開發，例如體外 ELAD 裝置。 此外，Hep-G2/C3A 可能易受某些人類病毒（例如茲卡病毒）感染，使其在以病毒學為重點的肝細胞系統研究中具有實用價值。

37°C，5% CO_₂，加濕環境。

1. Confirm that the vial remains deeply frozen upon delivery, as cells are shipped on dry ice to maintain optimal temperatures during transit.

2. Upon receipt, either store the cryovial immediately at temperatures below -150°C to ensure the preservation of cellular integrity, or proceed to step 3 if immediate culturing is required.

3. For immediate culturing, swiftly thaw the vial by immersing it in a 37°C water bath with clean water and an antimicrobial agent, agitating gently for 40-60 seconds until a small ice clump remains.

4. Perform all subsequent steps under sterile conditions in a flow hood, disinfecting the cryovial with 70% ethanol before opening.

5. Carefully open the disinfected vial and transfer the cell suspension into a 15 ml centrifuge tube containing 8 ml of room-temperature culture medium, mixing gently.

6. Centrifuge the mixture at 200 x g for 5 minutes, carefully discard the supernatant containing freezing medium.
