BC-3C

BC3c 是一種人類膀胱癌細胞系，源自一名成年女性受贈者的侵入性實體移行細胞癌手術活檢組織。該細胞系是透過酶解分離及在添加胎牛血清的 McCoy’s 5A 培養基中進行連續傳代，從原發性腫瘤組織建立而來。 BC3c 細胞呈現上皮形態，具有黏附生長特性，並能在體外形成凝聚性集落。免疫細胞化學分析顯示，該細胞表達細胞角蛋白 8 和 19，證實其上皮起源；而維曼汀及成纖維細胞相關抗原等間質標記物則未見表達。 該細胞不表達α-胎蛋白或癌胚抗原。細胞遺傳學分析顯示其具有近三倍體的核型，並伴有多種結構性及數目性染色體異常，這與侵襲性尿路上皮癌典型的基因組不穩定性相符。

分子分析未檢測到 H-ras 第 12 密碼子或 K-ras 第 12 及 13 密碼子的活化突變，且在 TP53 已檢測的熱點密碼子中亦未發現突變。 此外，免疫細胞化學法未檢測到 p53 蛋白的積聚，這表明該模型中不存在常見的 p53 穩定化事件。BC3c 細胞在體外展現出快速增殖，包括在低血清條件下的生長，這表明存在自主分泌的生長刺激。 來自 BC3c 培養的條件培養基可促進增殖，這支持了內源性生長促進因子存在的觀點。

Accutase 20 分鐘 37°C；

1. Confirm that the vial remains deeply frozen upon delivery, as cells are shipped on dry ice to maintain optimal temperatures during transit.

2. Upon receipt, either store the cryovial immediately at temperatures below -150°C to ensure the preservation of cellular integrity, or proceed to step 3 if immediate culturing is required.

3. For immediate culturing, swiftly thaw the vial by immersing it in a 37°C water bath with clean water and an antimicrobial agent, agitating gently for 40-60 seconds until a small ice clump remains.

4. Perform all subsequent steps under sterile conditions in a flow hood, disinfecting the cryovial with 70% ethanol before opening.

5. Carefully open the disinfected vial and transfer the cell suspension into a 15 ml centrifuge tube containing 8 ml of room-temperature culture medium, mixing gently.

6. Centrifuge the mixture at 200 x g for 5 minutes, carefully discard the supernatant containing freezing medium.

7. Follow the procedure described under Post-Thaw Recovery
