A9 細胞是一種源自小鼠脂肪組織的成纖維細胞樣細胞系。該細胞系由 W. R. Earle 於 1940 年以 L929 親本株的亞克隆形式建立。該親本株取自一隻雄性 C3H/An 小鼠的正常皮下疏鬆組織及脂肪組織。

這些細胞的一大顯著特徵在於其會表達腺苷磷核糖轉移酶（APRT）和次黃嘌呤磷核糖轉移酶（HPRT），因此被標示為 APRT+ 和 HPRT+。 這些細胞在病毒研究中具有重要價值，特別是涉及偽狂犬病毒（PRV）、印第安納株水疱性口炎病毒（VSV）以及單純疱疹病毒（HSV）的研究。

這種獨特的組合使 A9 細胞同時具備 B 淋巴細胞與骨髓瘤細胞的特性。總體而言，A9 細胞是一種成熟的成纖維細胞樣細胞系，廣泛應用於研究病毒感染（尤其是豬繁殖與呼吸綜合症病毒、水痘帶狀疱疹病毒及人類疱疹病毒）以及免疫學領域。

37°C，5% CO_₂，加濕環境。

1. Confirm that the vial remains deeply frozen upon delivery, as cells are shipped on dry ice to maintain optimal temperatures during transit.

2. Upon receipt, either store the cryovial immediately at temperatures below -150°C to ensure the preservation of cellular integrity, or proceed to step 3 if immediate culturing is required.

3. For immediate culturing, swiftly thaw the vial by immersing it in a 37°C water bath with clean water and an antimicrobial agent, agitating gently for 40-60 seconds until a small ice clump remains.

4. Perform all subsequent steps under sterile conditions in a flow hood, disinfecting the cryovial with 70% ethanol before opening.

5. Carefully open the disinfected vial and transfer the cell suspension into a 15 ml centrifuge tube containing 8 ml of room-temperature culture medium, mixing gently.

6. Centrifuge the mixture at 300 x g for 3 minutes to separate the cells and carefully discard the supernatant containing residual freezing medium.

7. Gently resuspend the cell pellet in 10 ml of fresh culture medium. For adherent cells, divide the suspension between two T25 culture flasks; for suspension cultures, transfer all the medium into one T25 flask to promote effective cell interaction and growth.

8. Adhere to established subculture protocols for continued growth and maintenance of the cell line, ensuring reliable experimental outcomes.
