SiHa

SiHa 細胞是一種人類子宮頸鱗狀細胞癌細胞系，數十年來廣泛應用於研究領域。該細胞系是從一名 55 歲日本女性鱗狀細胞癌患者的原發性子宮活檢組織片中分離出來的。 由於其獨特的遺傳特徵，該細胞系對研究子宮頸癌及其他相關疾病的研究人員極具吸引力。

研究發現，SiHa 細胞會表達 p53+ 和 pRB+ 基因，這些基因參與細胞週期調控、DNA 修復及腫瘤抑制。這些基因使 SiHa 細胞成為研究癌症發生與進展分子機制的理想模型。 此外，SiHa 細胞是合適的轉染宿主，使其成為基因表達研究的絕佳工具。

SiHa 細胞具有超三倍體核型，平均染色體數介於 69 至 72 之間。SiHa 細胞為 HPV-16 陽性，每細胞內整合有 1 至 2 份病毒基因組。 該細胞具有致瘤性，可在裸鼠體內形成分化程度低（III 級）的鱗狀細胞癌。這使其成為研究癌症進展及測試抗癌藥物之絕佳模型。

SiHa 細胞系可表達多種同工酶，包括 AK-1、ES-D、G6PD、GLO-I、Me-2、PGM1 及 PGM3。電子顯微鏡觀察顯示，細胞質中富含張力絲，細胞連接處則有橋粒。 SiHa 細胞屬黏附型細胞，在含 10% 胎牛血清 (FBS) 的培養基中，其倍增時間為 17 小時；在含 5% FBS 的培養基中，則為 21 小時。 92% 的 SiHa 細胞表現上皮細胞黏附分子（EpCAM），顯示其上皮起源。這些細胞表現出強烈的細胞角蛋白表達，但不表達維曼汀。

1. Confirm that the vial remains deeply frozen upon delivery, as cells are shipped on dry ice to maintain optimal temperatures during transit.

2. Upon receipt, either store the cryovial immediately at temperatures below -150°C to ensure the preservation of cellular integrity, or proceed to step 3 if immediate culturing is required.

3. For immediate culturing, swiftly thaw the vial by immersing it in a 37°C water bath with clean water and an antimicrobial agent, agitating gently for 40-60 seconds until a small ice clump remains.

4. Perform all subsequent steps under sterile conditions in a flow hood, disinfecting the cryovial with 70% ethanol before opening.

5. Carefully open the disinfected vial and transfer the cell suspension into a 15 ml centrifuge tube containing 8 ml of room-temperature culture medium, mixing gently.

6. Centrifuge the mixture at 300 x g for 3 minutes to separate the cells and carefully discard the supernatant containing residual freezing medium.

7. Gently resuspend the cell pellet in 10 ml of fresh culture medium. For adherent cells, divide the suspension between two T25 culture flasks; for suspension cultures, transfer all the medium into one T25 flask to promote effective cell interaction and growth.

8. Adhere to established subculture protocols for continued growth and maintenance of the cell line, ensuring reliable experimental outcomes.

若需長期保存，請將試管置於溫度約為 −150 至 −196 °C 的氣相液氮中。僅在轉移至液氮之前，可將試管暫時存放於 −80 °C 環境中，作為短期過渡步驟。

冷凍保存的細胞系會以乾冰冷藏，並裝於經驗證的隔熱包裝中，包裝內含有足夠的冷媒，以確保在運輸過程中溫度維持在約 −78 °C。收到貨物後，請立即檢查容器，並盡快將試管轉移至適當的儲存場所。
